冷冻大熊猫初乳XO和PAO活性测定及其与牛、羊乳的比较

摘    要:黄嘌呤氧化酶(XO)和多胺氧化酶(PAO)是乳汁中两类重要的氧化还原酶,与新生幼崽免疫系统的形成密切相关。采用牛、羊鲜乳及其冷冻乳建立XO和PAO活性的Amplex Red荧光检测方法,对冷冻大熊猫初乳中XO和PAO活性进行测定,并与新鲜和冷冻牛、羊乳中这两种酶的活性进行比较。结果表明,3 种乳中XO活性高于PAO活性;XO活性总体趋势为牛乳>大熊猫初乳>羊乳;PAO活性总体趋势为牛乳>羊乳>大熊猫初乳。大熊猫初乳通过XO、PAO和乳过氧化物酶LPO系统的协同抗菌机制在新生幼崽肠道微生物和先天免疫系统形成过程中发挥作用且以XO为主。

Abstract:Xanthine oxidase(XO) and Polyamine oxidase (PAO) are two important oxidoreductases in milk,which are closely related to the formation of the immune system in newborns.Amplex Red fluorescence assay for establishing XO and PAO activities by using fresh and frozen cow and goat milk was used to determine the activities of XO and PAO in frozen giant panda colostrum,and it was also compared with XO and PAO in fresh and frozen cow and goat milk.The results showed that the XO activities were higher than PAO activities in the three kinds of milk.The XO activities of the three kinds of milk were cow milk,giant panda colostrum and goat milk from order of high to low. The overall trend of PAO activities from high to low were cow milk,goat milk and giant panda colostrum. The synergistic antibacterial mechanism of XO,PAO and LPO systems in giant panda colostrum is dominated by XO,which plays a role in the formation of intestinal microorganism and innate immune system in newborn cubs.
