摘 要: | 目的]建立HPLC测定小麦中阿魏酸含量的检测方法。方法]色谱柱为Hyperclone BDS C18柱(150 mm×4.60 mm×5μm,phe-nomenex);流动相为甲醇酸性水溶液(V甲醇∶V水∶V冰乙酸=25∶75∶0.75);流速为1.0 ml/min;进样量为20μl;检测波长为320 nm;柱温为30℃。结果]阿魏酸进样浓度在80.8~242.4μg/ml范围内;在选定色谱条件下阿魏酸线性关系良好。结论]该方法操作简单,样品出峰时间较其他方法快,可应用于小麦中阿魏酸含量的测定。

Abstract: | Objective] To establish a HPLC method for the determination of ferulic acid in wheat.Method] The HPLC conditions were: Hyperclone BDS C18(150 mm×4.60 mm×5 μm,phenomenex),mobile phase of carbinol acidic water(Vcarbinol∶ Vwater∶ Vglacial acetic acid=25∶ 75∶ 0.75),flow rate of 1.0 ml/min,sampling amount of 20 μl,detection wavelength of 320 nm and column temperature of 30 ℃.Result] In the range of 80.8-242.4 μg/ml,the relationship between concentration of quercetin and peak area was good(r=0.999 7).Conclusion] By comparison with other methods,the method proposed in this paper has the characters of quick peak flowing out.This method can be used to determine ferulic acid in wheat. |
