| 摘 要： | [目的]克隆地衣芽孢杆菌BL03的脂肪酶基因LIP并对其序列进行分析。[方法]以地衣芽孢杆菌BL03基因组核酸为模板,PCR扩增得到地衣芽孢杆菌脂肪酶基因,用pMD19-T载体克隆后进行PCR鉴定及序列测定和生物信息学分析。[结果]测序结果表明该片段完整开放阅读框大小为615 bp,与已发表的地衣芽孢杆菌LTP基因(GeneID:3098655)相比,核酸序列相似为99.67%;推导编码氨基酸序列相似性为99.02%。[结论]该基因的获得为其异源表达及相关研究奠定了基础。

| Abstract: | [Objective] To clone Bacillus licheniformis BL03 gene LIP,and analyze the sequence.[Method] The LIP gene encoding lipase from B.licheniformis BL03 was amplified by PCR and cloned to the vector pMD19-T.[Result] Nucleotide sequence analysis of the LIP gene revealed that the coding region was formed of one complete ORF including 615 base pairs.The cloned gene was 99.67% similar with B.licheniformis LIP gene(GeneID:3098655) and the putative amino acid sequence was 99.02% similar respectively.[Conclusion] The cloned gene laid a foundation for the heterologous expression and relevant researches. |
