竹叶多糖的提取及抗氧化作用研究

摘    要:目的]研究竹叶多糖的抗氧化作用。方法]从提取竹叶黄酮后的残渣中提取竹叶多糖。用抑制Fenton反应产生·OH使溴甲酚紫褪色法测定竹叶多糖清除·OH的作用,以硫脲为对照。用清除DPPH法测定竹叶多糖清除自由基的作用,以Vc为对照。结果]竹叶多糖的提取率为1.15%;纯度达到83.33%。竹叶多糖的剂量从0.01 mg增加到0.08 mg,对·OH清除率从16.97±0.87(%)增加到63.89±0.66(%);剂量再增加到0.09 mg,清除率不再增加。硫脲的剂量从0.01 mg增加到0.08 mg,对·OH清除率从13.89±4.17(%)增加到91.62±6.30(%);剂量增加到0.09 mg时,清除率也不再增加。剂量从0.02 mg到0.09 mg,硫脲对·OH的清除作用均显著高于同剂量竹叶多糖。竹叶多糖的剂量从0.01 mg增加到0.75 mg,对DPPH的清除率从11.56±1.13(%)增加到73.66±0.28(%),剂量增加到1.0 mg时,清除率不再增加;Vc的剂量从0.01 mg增加到0.5 mg,对DPPH的清除率从18.19±0.40(%)增加到89.52±0.28(%),剂量增加到0.75 mg时,清除率也不再增加;Vc的作用均显著高于同剂量的竹叶多糖。结论]竹叶多糖对清除·OH和DP-PH有较好的作用。

Abstract:Objective] To study the antioxidant functions of polysaccharide from bamboo leaf.Method] After flavonoids were extracted,the polysaccharide was extracted from bamboo leaf.Function of removing ·OH by polysaccharide from bamboo leaf was determined with Fenton method producing ·OH and bromcresol purple was faded with thiourea as the control.Function of removing free radicals was determined with method of removing DPPH,the control was Vc.Result] The extraction rate of polysaccharide was 1.15%,purity of polysaccharide was 83.33%.The dose of polysaccharide increased from 0.01 mg to 0.08 mg,the clearance to ·OH increased from 16.97±0.87(%) to 63.89±0.66(%),but dose increased to 0.09mg,the clearance will not increase.Dose of thiourea increased from 0.01 mg to 0.08 mg,the clearance to ·OH increased from 13.89±4.17(%)to 91.62±6.30(%).When dose increased to 0.09 mg,the clearance will not increase,and dose of thiourea increased from 0.02 mg to 0.09 mg,its function was marked higher than the same polysaccharide.The dose of polysaccharide increased from 0.01 mg to 0.75 mg,clearance to DPPH increased from 11.56±1.13(%)to 73.66±0.28(%),but dose increased to 1.0 mg,clearance will not increase.Dose of Vc increased from 0.01mg to 0.5 mg,clearance to DPPH increased from 18.19±0.40(%) to 89.52±0.28(%),when dose increased to 0.75 mg,the clearance will not increase,and its function was higher than the same polysaccharide markedly.Conclusion] The polysaccharides from bamboo leaf have remarkable function of removing ·OH and DPPH in vitro.
