摘    要:为保证葡萄白腐病生防菌瘦果红酵母液体培养的菌体数,采用单因素和正交设计的方法对瘦果红酵母增殖培养基组分进行了优化。首先通过单因素试验确定了适宜瘦果红酵母生长的碳源、氮源、碳源浓度、氮源浓度和无机盐种类,再通过L9(34)正交试验进一步确定了瘦果红酵母J1增殖培养基的最佳配方为:葡萄糖3%、牛肉膏1%、豆饼粉0.5%、K2HPO40.03%、MgSO40.05%。利用该培养基,在100mL三角瓶中,28℃、150r/min振荡培养56h,其菌数可达15.58×107cfu/mL。

Abstract:Rhodotorula acheniorum is a biological control fungus for grape white rot.To obtain enough quantity of the fungus,liquid culture medium was optimized by the method of single factor and orthogonal experimental design.Carbon source,nitrogen source,concentration of carbon source,concentration of nitrogen source and inorganic salt were determined with single factor experimental design.Then L9(34)orthogonal experimental design was used to determined the optimal medium component for Rhodotorula acheniorum J1.The optimal medium component was 3% of glucose,1% of beef extract,0.5% of bean cake powder,0.03% of K2HPO4,and 0.05% of MgSO4.With this medium,the number of Rhodotorula acheniorum reached 15.58×107 cfu/mL after shaking with 150r/min at 28℃,in 100mL conical flask for 56h.
