植物透射电镜样品制备技术Improvement and comparison with different resins of madding ultrathin section for the transmission electron microscope specimen-making的改进及不同树脂选择比较

摘    要:针对植物细胞壁组织坚硬,透射电镜样品制备难度大的问题,笔者对样品制备程序化进行了探讨;同时,在样品制备过程中对不同树脂配方进行了选择和比较,结果表明:在经优化的样品处理下,不同树脂均能观察到植物样品的超微结构,结构清晰,且环氧树脂618包埋的样品受环境影响较小。In view of the hard plant cell wall, there is some problem during the transmission electron microscope (TEM) sample preparation. The authors probed into procedural sample preparation, at the same time, different resins were selected and compared. The results showed that ultra-microstructure were observed distinctly using different kinds of resin under the same conditions, the epoxy resin 618 is less affected by the environment in the process of embedding.
