不同梓属树种花器官蛋白电泳分析

摘    要:为了分析与楸树自交不亲和有关的特异蛋白质特性,采用考马斯亮蓝染色法和SDS-PAGE技术研究了2种梓属树种(滇楸、楸树)花器官的蛋白质含量及蛋白质组分。结果表明:楸树-1(连云港种源)的花柱、子房、花萼蛋白质含量较低,滇楸和楸树-2(老山种源)较高,而楸树-1花瓣蛋白质含量略高于其他树种;同一树种不同器官蛋白质含量表现为子房>花萼>花柱>花瓣。蛋白组分分析表明,梓属各树种不同花器官内的蛋白质主要集中在12~45 ku之间,各器官共有蛋白20个左右,发现具有特异性的蛋白包括:楸树-1花柱42 ku蛋白,楸树-2花柱64和19 ku蛋白,滇楸花柱37 ku蛋白,以及楸树-2和滇楸花柱共有的58和24 ku蛋白;楸树-1子房41、38和23 ku蛋白;花萼中,88和65 ku蛋白为楸树-1所有,52、40、26 ku蛋白为楸树-2和滇楸特有。花瓣内蛋白带较少,仅发现63 ku蛋白为楸树-1特异蛋白。

Abstract:To understand the mechanisms of self-incompatibility of Catalpa genus and its relationship with the specific proteins,the protein fractions and soluble proteins contents in the different floral organs of the three tree species of Catalp genus [including C.fargesii,CB-1(C.bugeicorne come from Lianyun),CB-2(C.bugeicorne come from Laoshan)] were studied by the techniques of SDS-PAGE and Brilliant Blue and Commassie Dye.The results showed that the soluble proteins contents in the style,ovary and calyx of CB-1 were lower than that of C.fargesii and CB-2,while the soluble proteins contents in the petal of CB-1 were higher than that of the other species.For each of three species,the soluble proteins contents in the organs were in the order at ovary>calyx>style>petal.There were 20 proteins bands observed in the floral organs of three species and the MW(relative molecular weight) were mainly from 12 to 45 ku.By the techniques of SDS-PAGE,some specific proteins were found in the different floral organs of the species.In the style,the 42 ku protein was found in CB-1,the 64 and 19 ku proteins in CB-2,and the 37 ku protein in C.fargesii.The 58 and 24 ku proteins were found both in CB-2 and in C.fargesii.The 41,38 and 23 ku proteins were found in the ovary of CB-1.In the calyx,the 88 and 65 ku proteins were only found in CB-1,and the 52,40 and 26 ku proteins was found in CB-2 and C.fargesii.While in the petal,only 63 ku protein was found in CB-1.
