摘    要:[目的]优化大叶白麻茎多酚的提取工艺,并考察其体外抗氧化性。[方法]采用响应面法对大叶白麻茎多酚的提取工艺进行了优化,考察了料液比、超声强度、超声时间对提取得率的影响,以V_C为参照,将大叶白麻茎多酚和茶多酚的抗氧化性进行对比。[结果]大叶白麻茎多酚的最佳提取工艺是料液比1∶17,超声强度410 W,提取时间60 min,在该条件下大叶白麻茎多酚的提取得率为2.09%,提取时间和超声强度对提取得率的影响均为极显著。还原力与还原剂浓度之间有较好的量效关系,三者的还原力从高到低依次为大叶白麻茎多酚、茶多酚、V_C,与抗氧化能力的强弱性一致。大叶白麻茎多酚清除羟基自由基的能力是茶多酚的1.34倍,清除超氧阴离子、DPPH·自由基的能力与茶多酚较为接近,且明显高于V_C,分别是V_C的2.00、2.80倍。[结论]大叶白麻茎多酚具有良好的抗氧化活性,应加强对其进一步的应用研究。

Abstract:[Objectuve] The aim of the study was to optimize extraction technology of polyphenol in stem of Poacynum hendersonii and study its antioxidation in vitro.[Method]The extraction technology of polyphenol was optimized by response surface analysis and the effect of liduid ratio,extraction time and ultrasonic intensity on extraction yield of polyphenol was investigated.VC taken as reference,antioxidant activity of polyphenol in Poacynum hendersonii and tea were compared and analyzed.[Result] The optimal extraction technology was as follows solid-liquid ratio of 1∶17,ultrasonic intensity of 410 W,extraction time for 60 min,under this technology,the extraction yield of polyphenol from Poacynum hendersonii was 2.09%.Both influence of time and ultrasonic intensity on the yield was very significant.The order of antioxidative effect from high to low was polyphenol in Poacynum Hendersonii,polyphenol in tea,VC.As the dose of the three antioxidant increasing,reducing power strengthens,displays good does-effect relationship.The order of reducing power and antioxidative effect were identical.Hydroxyl radical scavenging ability of polyphenol in Poacynum Hendersonii was about 1.34 times of polyphenol in tea.DPPH· and superoxide anion scavenging ability of polyphenol in stem of Poacynum hendersonii were close to that in tea,2.00,2.80 times of VC.[Conclusion]The polyphenol in stem of Poacynum hendersonii has been shown to exhibit excellent antioxidant activity,so it should strengthen the further application research.
