摘    要:目的]查找水稻RSSG58在拟南芥中的同源基因并观察其组织表达模式。方法]利用相关基因信息学网站构建水稻RSSG58的基因进化树,构建其同源基因ATG58::GUS定位载体转入拟南芥,观察分析ATG58的组织表达模式。结果]进化树和序列对比发现ATG58与RSSG58具有较高的序列相似性,GUS定位遗传材料显示ATG58在多种组织都有表达,在生长发育早期的根、子叶、下胚轴、胚芽表达量相对较高,在生殖发育阶段的花苞和花药内优势表达,ATG58与RSSG58具有相似的GUS定位组织表达模式。结论]ATG58是水稻RSSG58的同源基因,二者组织表达模式一致,这些结果为深入研究拟南芥ATG58和水稻RSSG58在雄配子发育过程中的作用及机制提供了基础资料。

Abstract:Objective] To find the homologous gene of rice RSSG58 in Arabidopsis thaliana and observe its tissue expression pattern.Method] The phylogenetic tree of rice RSSG58 was constructed by using the related gene informatics website.The homologous gene ATG58::GUS locus was constructed into A.thaliana to observe and analyze the tissue expression pattern of ATG58.Result] The phylogenetic tree analysis and sequence comparison showed that ATG58 had a highest sequence similarity with RSSG58.GUS locating genetic material showed that ATG58 was expressed in various tissues.The expression of ATG58 was relatively higher in root,cotyledon,hypocotyl and embryo of seedling stage,and it was superior to the buds and anthers in the reproductive development stage.ATG58 had similar GUS localization expression pattern with RSSG58.Conclusion] ATG58 is the homologous gene of rice RSSG58,which is consistent with RSSG58 in the tissue expression pattern.These results provide a basis for further study of the role and mechanism of A.thaliana ATG58 and rice RSSG58 in the development of male gametes.
