摘    要:目的]对贵州省赤水河流域茅台段土壤中放线菌进行分离、培养与DNA提取。方法]从赤水河流域采集土壤样本,采用土壤稀释分离法和划线纯化相结合的方法进行放线菌分离与培养,并对其进行形态学观察。利用CTAB法提取放线菌基因组DNA。结果]共分离到6株放线菌。0.001 g/mL土壤悬液最适宜分离放线菌。提取的放线菌DNA条带完整,大小约为23.13 kb。结论]该研究结果可为放线菌的进一步开发利用提供试验材料和理论依据。

Abstract:Objective] To isolate and culture actinomycetes in the soil from Maotai Section of Chishui River Basin in Guizhou to make DNA extraction.Method] Soil samples were collected from Chishui River Basin to isolate and culture actinomycetes by using dilution method and scraping line method.And their morphological characteristics were observed.The genomic DNA was extracted from actinomycetes by using CTAB method.Result] 6 strains of actinomycetes were isolated.The suitable concentration of soil suspension for the isolation of actinomycetes was 0.001 g/mL.The extracted DNA from actinomycetes had complete bands with the size of about 23.13 kb.Conclusion] The research results can provide test materials and theoretical basis for further exploitation and utilization of actinomycetes.
