甜叶菊NaN3离体诱变技术体系的建立

摘    要:采用NaN3对甜叶菊离体培养的茎段进行诱变处理,通过对各处理下的再生苗进行SRAP检测,建立其离体诱变技术体系。结果显示,较低浓度的NaN3(3 mg/L)诱变处理对再生苗伤害较小,随着诱变浓度的增加(3~7 mg/L),茎段死亡率也逐渐增加;SRAP分子检测发现,再生苗的DNA序列发生了不同程度的多态性变化,最高多态性比率为54.2%(3 mg/L处理20 d)。结果表明,较合适NaN3离体诱变技术体系为3 mg/L处理20 d。

Abstract:The stems of Stevia rebaudiana Bertoni were used as the explants for in vitro mutation inducted by NaN3, and the system of mutation in vitro was established by investigating the new plantlets with SRAP technology. The results indicated that, little harm appeared in the stems by low concentration of NaN3(3 mg/L), and the death rate of stems was improved with the increase of NaN3 concentration(3~7 mg/L). Meanwhile, DNA sequence of regenerated plantlets was changed by SRAP analysis, with the highest mutation ratio of 54.2% in the treatment of 3 mg/L NaN3 for 20 days. Based on the results, the optimal system for mutation was stems of S. rebaudiana treated by 3 mg/L of NaN3 for 20 days.
