天津市小麦田荠菜对双氟磺草胺的抗性及其乙酰乳酸合成酶基因突变

摘    要:为明确天津市小麦田荠菜种群对双氟磺草胺的抗性情况及可能的抗性机理,本研究在天津市静海区、武清区、宝坻区及蓟州区等荠菜发生严重地区的小麦田共采集到6个荠菜种群,采用整株水平测定法测定了6个荠菜种群对双氟磺草胺的抗性水平,并扩增、比对了其靶标乙酰乳酸合成酶 (ALS) 基因部分片段的差异。结果表明:6个荠菜种群对双氟磺草胺均产生了高抗性,抗性倍数在11.4~47.2之间。对抗性和敏感种群的ALS基因片段进行测定比对发现,6个荠菜种群ALS基因197位氨基酸均由脯氨酸 (CCT) 突变为丝氨酸 (TCT),该突变可能是导致荠菜种群对双氟磺草胺产生抗性的重要原因之一。

Resistance to florasulam and mutation of the acetolactate synthase gene of Capsella bursa-pastoris in winter wheat fields in Tianjin

Abstract:In order to examine the resistance level and the potential resistance mechanism of Capsella bursa-pastoris to florasulam, six populations from wheat fields at Jinghai, Wuqing, Baodi and Jizhou in Tianjin were collected. Whole-plant bioassay was conducted to investigate the florasulam resistance levels, and acetolactate synthase (ALS) gene fragments were amplified, sequenced and compared between the resistant and susceptible populations. The results showed that the six populations were highly resistant to florasulam, and the resistance indexes were between 11.4 and 47.2. These six resistant populations were found to contain one amino acid mutation of proline (CCT) to serine (TCT) at the codon position 197 in the ALS gene. This mutation in the ALS gene might be one of main reasons for the resistance to florasulam in Capsella bursa-pastoris populations.
