eNOS基因转染对大鼠颈总动脉损伤后再狭窄程度的影响

摘    要:为了研究eNOS基因转染对大鼠颈总动脉损伤后再狭窄程度的影响,以2F导管制作球囊损伤的大鼠颈总动脉内膜剥脱模型。用重组真核表达载体pcDNA3.1-eNOS经Fugene6介导体内转染大鼠的损伤后血管平滑肌细胞,2w、1m、2m后处死大鼠,取出大鼠的左侧颈总动脉及对侧正常的颈总动脉,分别应用HE染色和Verhoeff铁苏木精染色,观察血管再狭窄的程度,结果发现:体内eNOS转染组的血管内膜增生明显减弱,再狭窄程度显著低于体内空白对照组和空载pcDNA3.1(-)转染组(P<0.01)。而后二者之间无显著差异(P>0.05)。

Abstract:To study the effect of eNOS gene transfection on the extent of re-s tricture of the common carotid artery in rat after injury, an animal model with intimal stripping of the common carotid artery after injury of the saccule was e stablished by using a 2F catheter. The recombinant pcDNA3.1-eNOS vector was use d to in vivo transfect the rat vascular smooth muscle cells mediated by Fugene 6 . The rats were sacrificed after 2 weeks, 1 month and 2 months, respectively. Th e left common carotid artery and the right normal common carotid artery were har vested, HE and Verhoeff Fe hematoxylin stain were performed to evaluate the exte nt of re-stricture of the vessels. The results showed that in vivo transfection of eNOS gene could inhibit the intimal proliferation significantly. The extent of vascular re-stricture in the experiment group was significantly lower than t hat seen in the blank control and blank transfection (without pcDNA3.10 group (P <0.01). There was no significant difference between the blank control and blank transfection group(P >0.05).
