龟裂链霉菌菌株MY02发酵液中抑茵活性组份SN06的分离纯化

本实验室自蔬菜保护地筛选的一株链霉菌菌株,经鉴定为Streptomycesrimosus daheishanensis,编号为MY02,其发酵液中活性组份SN06对多种蔬菜真菌病害都有较好的防治效果.采用Betina溶剂系统进行了SN06薄层层析展开剂的筛选,结果表明,活性组分SN06在弱极性的展开剂中分离效果较好,可选用正丁醇的水饱和溶液、甲醇:水(40:60)、正丙醇:水(40:60)作为适宜的展开剂.采用pH层析和纸电泳方法分析SN06的电离性质,结果表明,SN06属于中性抗生素.在此基础上,确定选用萃取法初步分离SN06,选用的最佳萃取剂为正丁醇,最后采用硅胶薄板层析法,得到活性组分SN06淡黄色晶体.经高压液相色谱分析,分离提取的活性组分SN06纯度可达96%.

Purification of active components SN06 in fermentation of Streptomyces rimosus MY02

Streptomyces rimosus daheishanensis was scanned from protected vegetable field.Ac-tive components SN06 extracted from fermentation of Streptomyces rimosus daheishanensisshowed antagonism against pathogenic fungus such as Fulvia fulva,Botrytis cinerea.Polarity,dissolution and ionization characteristics have been analysed.The developing solvent wasscreened by Betina paper chromatography system,the result showed that n-butanol solution sat-urated with distilled water,methanol:distilled water(40:60),n-propyl alcohol:distilled wa-ter(40:60)is the good developing solvent.SN06 is a neutral antibiotic by pH-PC and paper e-lectrophoresis.At first the fermentation was filtered and extracted with n-butanol,and then theslight yellow crystal of SN06 obtained by silica gel separation.Analysis of HPLC showed thatthe purity of SN06 is 96%.
