活性炭促进烟草花药培养出苗的作用机制研究

摘    要:本文以Nitsh H(1967)为基本培养基,烟草花药为培养材料,研究了活性炭影响烟草花药培养出苗率的作用及机制。结果是:在一定范围内,培养基中大量元素与微量元素的比例比绝对含量对出苗率的影响大;活性炭对烟草花药培养出苗的促进作用受培养基中大量元素、微量元素平衡程度的影响;活性炭试剂级别不同对花培出苗率的影响不同;活性炭中促进出苗的物质部分可以被EDTA提取出来;培养基中附加Zn、Fe、P、K等元素能不同程度的促进出苗。结果说明:培养基中元素平衡对烟草花药培养胚状体的形成极为有利;活性炭加入培养基,通过平衡培养基中元素比例和增加促进出苗的微量元素而促进烟草花药培养胚状体的形成。

Abstract:Used the basic culture medium Nitsh H(1967) and tobacco anther to studying the mechanism of AC promoting tobacco anther culture emerence. It was a great influence to rate of emergence which the proportional absolute content of macroelement and microelement in culture medium. AC's promotion of tobacco anther culture emergence was effected with macroelement and microelement's balance level. When AC reagent grade was different, rate of emergence of anther culture isn't alike, either. AC's matter portion of promoting emergence could be distilled by EDTA. And when culture medium was subjoined Zn, Fe, P, K, rate of emergence had a different digree's increase. So it can be confirmed that culture medium's element balance was benefical to tobacco anther culture embryoid's forming. And yet, when AC subjoin in culture medium, which by adjust element proportion and increasing microelement of promoting emergence, the embryoid's from was hastened,too.
