摘 要: | 以铁线莲Gipsy Queen的茎节和茎段为外植体,在MS基本培养基中添加0、1.0、2.0 mg/L 6-BA 和0、0.01、0.05 mg/L NAA进行初代诱导和继代增殖培养。结果表明院最适合诱导愈伤组织的培养基为MS+NAA 0.01 mg/L+6-BA2.0 mg/L,诱导率可达50.0%,诱导出的愈伤组织呈淡黄色疏松颗粒状;最适合诱导侧芽的培养基为MS+NAA 0.01mg/L+6-BA 1.0 mg/L,出芽率可达170%;最适增殖培养基为MS+NAA 0.01 mg/L+6-BA 2.0 mg/L,增殖倍数达3.39。

Abstract: | Callus and adventitions bud of Gipsy Queen were induced with columella and stem of Gipsy Queen asexplants and with the MS added different concentrations of 6-BA (0, 1.0, 2.0 mg/L) and NAA (0, 0.01, 0.05 mg/L) asnutrient medium. The results showed that the callus could be easily induced in MS+NAA 0.01 mg/L+6-BA 2.0 mg/L, theinduction rate was 50.0%, and the induced callus was loose and granular appeared yellowish. The best lateral bud inducedmedium was MS+NAA 0.01 mg/L+6-BA 1.0 mg/L, and the budding rate was 170%. The best multiplied medium was MS+NAA 0.01 mg/L+6-BA 2.0 mg/L, and the proliferation times was up to 3.39. |
