摘    要:根据GenBank中DuCV基因序列,在保守区设计了6条特异性引物,并对反应条件进行优化,建立了一种适用于鸭圆环病毒(DuCV)的环介导等温扩增快速检测方法(LAMP)。该方法对H9亚型禽流感、小鹅瘟、鸭瘟、鸭肝炎、鸭副黏病毒均无扩增反应;且扩增反应只需在常规水浴锅中进行,1小时内即可完成反应;对DuCV模版DNA的最小检测限为10龟,灵敏度是一步法PCR的1000倍。本研究建立的LAMP方法简便、快速、灵敏、特异,适合在基层进行DuCV的快速检测。

Development of loop-Mediated Isothermal Amplification Assay for Visual Detection of Duck Circovirus

Abstract:A loop-mediated isothermal amplification (LAMP) assay was developed for detection of duck circovirus (DuCV) . According to the sequences of DuCV gene available in GenBank, six specific primers to gene were designed, and the reaction conditions were optimized. The results showed that the assay had no cross-reaction with H9 subtype AIV, gosling plague virus, duck plague virus, duck hepatitis virus, duckparamyxovirus and other grallatorial pathogens. The assay was performed in water bath within one hour and the amplification result was visualized by inspecting the white sediment and the green colour.. The detection limit of LAMP assay was 10 fg, 1 000-fold higher than that of the routine PCR. These results suggest that the LAMP assay is a simple and specific method for rapid detection of DuCV in field conditions.
