鳞球茎茎线虫实时荧光PCR检测技术的研究

摘    要:传统的线虫形态学鉴定存在经验性强和幼虫难以鉴定等不足.本文根据线虫ITS基因序列多态性,设计鳞球茎茎线虫(Ditylenchus dipsaci)特异的TaqMan探针,建立了实时荧光PCR检测方法.对茎线虫属的鉴定实验表明:只有鳞球茎茎线虫产生特异性荧光信号,同属的其他线虫均检测到荧光信号.整个检测过程只需30min到2h,能够满足口岸检验检疫快速通关的要求.

Abstract:Stem bulb nematode(Ditylenchus dipsaci )is a dangerous and damage nematodes in the world,and is also in the A 2 li st of quarantine pests concerned to China.A real time PCR method was developed for detection of D.dipsaci.The species-specific probe for D.dipsaci was developed,and no fluorescent signal was detected fo r other nematode except D.dipsaci.The detection process only n eeds 30min or 2 hours,so the method is specific and little time consumption.
