鸡免疫球蛋白G的提纯及定量测定

摘    要:从成年鸡血清中用硫酸铵三次沉淀粗提r球蛋白。然后,用G200凝胶柱过滤,洗脱液含O.14M NaCl,0.01M PB,流速6—8滴/分。取第二峰,用DE52纤维素柱层析,洗脱液是0.1M PH7.0 PBS。洗脱蛋白用免疫电泳等技术鉴定为纯化IgG。以此抗原1毫克加福氏完全佐剂首次免疫家兔后,再用同种抗原2—3次加强免疫,总蛋白用量3—5毫克。兔抗鸡IgG抗血清效价在1:16—1:32以上。以单向免疫扩散试验,测出不同周龄鸡血清中IgG的含量: 2周龄:2.9117±0.0779(n=20); 4周龄:3.8748±O.0591 (n=20); 10—13周龄:8.2243±O.4000 (n=59); 18—21周龄:13.3107±0.7434(n=44);


PURIFICATION AND QUANTITATIVE MEASUREMENT OF CHICKEN IMMUNOGLOBULIN G
Abstract:γ-globulin from chicken sera were extracted, and purified by gel filtration onsephadex G 200 column. The second peak portion was purified by DEAE-cellulosechromatical column. The collected specimen was identified through immunoelectrophoresis,double diffuse and polyacrylamide gel electrophoresis techniquesas purified IgG. Rabbits were immuned four times with 3—5 mg purified chicked IgG. Thetiter of anti—chicked IgG from rabbit ranged from 1: 16 to 1: 32. The IgG levels of White Leghorn of differeut week-ages(wk)were determinedas fellows: 2 wk: 2. 9117±0. 0779(n=20) 4 wk: 3. 8748±0. 0591(n=20)10—13wk: 8. 2243±0.4000(n=59)18—21wk: 13.3107±0.7434(n=44)Data obtained could be used for further experimental study.
