樱桃不同基因型再生植株的比较

摘    要:对甜樱桃(PrunusaviumL.)、杂种优系F系(P.avium L.×P.pseudocerasusL.)和H系(P.cerasus L× P.pseu-docerasusL.)等不同基因型的离体再生植株方式进行了比较研究。结果表明,不同遗传背景的樱桃试材,其再生方式和途径存在差异。其中甜樱桃品种美早的离体叶片在WPM+6-BA2.0mg·L-1+IAA2.0mg·L-1+GA30.1mg·L-1培养基上,不定芽再生率可达到90%,H10、萨米脱、F10、F8在6-BA(2~2.5mg·L-1)、IAA(1.0~2.0mg·L-1)的激素配比,也获得了76%、86%、80%、64%的再生率;杂种H10叶柄接种于改良MS+6-BA2.0mg·L-1+IBA0.1mg·L-1+2,4-D0.5mg·L-1的培养基,可以得到40%的再生率,其他品种仅有H8的叶柄获得了少量的再生植株;杂种F10的幼嫩节间接种于LS+6-BA1.0mg·L-1+NAA2.0mg·L-1+CH100mg·L-1培养基,最高再生率为40%,6-BA(1~2mg·L-1)和NAA(1~2mg·L-1)配比,H8、H10的离体节间再生率最高分别为25%和30%;杂种H8整体根在MS+6-BA1.0mg·L-1+2,4-D2.0mg·L-1+IBA0.05mg·L-1+NAA0.05mg·L-1的培养基上,再生率为96.7%,6-BA(0.5~2mg·L-1)和2,4-D(0.5~2.0mg·L-1)配比,H10、F8、F10离体根再生率分别为93.3%、96.7%和93.3%。

Comparative studies on plant regeneration from different genotypes of cherry plants
Abstract:The leaves, leaf stalk, internodes and root tips of the seedlings with different genetic background derived from Cerasus avium L.;C.avium L.× C.pseudocerasus L.were used as materials in vitro for studying their regeneration techniques.Results showed that seedlings with different genetic background had specific patterns and routes for plant regeneration.Satisfactory regeneration can be obtained in different materials by choosing special induction and culture manipulations.The primary results are listed briefly as follows:above 90% of in vitro leaves of sweet cherry cultivar Meizao regenerated adventitious buds on WPM+6-BA 2.0 mg·L-1(the same as next)+IAA 2.0 mg·L-1+GA3 0.1 mg·L-1, the best regeneration of seedling of H10, Summit, F10 and F 8 were 76%, 86%, 80% and 64% respectively in combinations of 6-BA(2 to 2.5 mg·L-1) and IAA(1.0 to 2.0 mg·L-1).When using leafstalks of hybrid H10 in vitro, 40% regeneration can be obtained on modified MS+6-BA 2.0 mg·L-1+IBA 0.1 mg·L-1+2,4-D 0.5 mg·L-1, a few regenerations were also observed on leafstalks of H8.LS medium supplemented with 6-BA and NAA was suitable for regeneration when taking young internodes as explants.In com-binations of 6-BA(1 to 2 mg·L-1)and NAA(1 to 2 mg·L-1), regeneration of H8 and H10 were 25% and 30% respectively, 6-BA 1.0 mg·L-1+NAA 2.0 mg·L-1 was optimal for regeneration of hybrid F10, 40% regeneration can be obtained.As high as 96.7% of intact root system of hybrid H8 regenerated buds on MS+6-BA 1.0 mg·L-1+2,4-D 2.0 mg·L-1+ IBA 0.05 mg·L-1+ NAA 0.05 mg·L-1.The best regeneration on intact root system of H10, F8 and F10 were 93.3%,96.7% and 93.3% respectively in combination of 6-BA(0.5 to 2 mg·L-1) and 2,4-D(0.5 to 2.0 mg·L-1).
