鸡黄胫基因BCDO2的多态性分析

摘    要:本研究设计PCR-RFLP方法,检测了农大隐性白C系、白来航、北京油鸡、三黄鸡、丝羽乌骨鸡、寿光鸡、菊花鸡、茶花鸡8个品种的BCDO2基因多态性,其中农大C系、白来航、北京油鸡均为GG纯合型,茶花鸡表现为AA纯合型;而丝羽乌骨鸡、寿光鸡和菊花鸡等品种的BCDO2基因的杂合度分别为0.5、0.4、0.13;本试验所用的三黄鸡品系为培育品系,也存在一定的杂合度,研究结果说明黄皮肤基因在我国多个地方品种还没有进行过选择。本研究还检测了BCDO2基因在三黄鸡和农大隐性白C系鸡种背部皮肤的表达量,结果显示,三黄鸡的BCDO2基因表达量高于农大C系,推测BCDO2基因的表达对背部皮肤的颜色仍起着重要的作用,并认为鸡躯干部皮肤与胫部皮肤色素的不对称分布主要由不同部位皮肤的组织特点和类胡萝卜素的化学性质决定。

Abstract:The BCDO2 gene polymorphism of eight chicken lines including the CAU-C recessive white,white Leghorn,Beijing you,three-yellow chicken,Silkie,Shouguang,Juhua and Chahua were detected on the A/G mutation.The results showed that CAU-C recessive white line,white Leghorn,Beijing you are fixed with G allele and Chahua had been fixed with a allele.While BCDO2 gene heterozygosity in the Silkie,Shouguang and Jvhua chicken is 0.5,0.4,0.13 respectively.The other breeds were polymorphism on this locus,indicating that these breeds were not under selection pressure of yellow skin color.The expression of BCDO2 gene on body skin of three-yellow and CAU-C recessive white was detected by real-time PCR.The results showed that the expression of BCDO2 gene in three-yellow chicken is higher than that in CAU-C recessive white but the difference did not reach significant level(P>0.05),this result implies an effect of BCDO2 gene on body skin color,but experimental repeats were still required to confirm.Meanwhile,the asymmetric distribution of carotenoids in the chicken trunk skin and shank skin is mainly determined by the skin characteristics of different tissue and the chemical properties of carotenoids.
