摘    要:通过超声波辅助农杆菌介导法,优化"美人指"葡萄遗传转化体系。利用GUS瞬时表达的方法研究了不同功率、时间条件下的超声波处理,外植体类型以及AS浓度对"美人指"葡萄遗传转化效率的影响。结果表明:外植体茎段在含75mg/L AS农杆菌悬浮液中侵染4 min,并在此期间,用功率为90 W的超声波处理14 s,获得最佳GUS瞬时表达率42.8%。

Abstract:In order to optimize the sonication-assisted Agrobacterium-mediated genetic transformation system of grape variety Meirenzhi,the effects of sonication treatment with different powers of ultrasonic for different time,different explant types and AS concentrations on the transformation efficiency were studied through the method of GUS instantaneous expression.The results showed that infecting explant stem segment with 75 mg/L AS suspension for 4 min,and in this period conducting sonication treatment with 90 W ...
