根瘤农杆菌生产琥珀酰多糖发酵条件的优化

摘    要:[目的]优化根瘤农杆菌产琥珀酰多糖的发酵培养条件。[方法]通过单因素试验和正交试验优化根瘤农杆菌产琥珀酰多糖的发酵培养条件。[结果]最终得到的优化培养条件为:蔗糖70 g/L,酵母粉6 g/L,CaCO_310 g/L,MgSO_41.5 g/L,KH_2PO_40.025 g/L,发酵时间4 d,种子接种量2.5%,培养基初始pH 7.2,发酵温度30℃,摇瓶转速250 r/min。在上述最优方案下,根瘤农杆菌琥珀酰多糖产量达18.550 g/L,比其初始条件下产量(8.010 g/L)明显提高。[结论]该研究为利用根瘤农杆菌生产琥珀酰多糖提供了理论依据。

Abstract:[Objective] The aim was to optimize the fermentation conditions for succinoglucan production by Agrobacterium tumefacien. [Method] We optimized the fermentation conditions for succinoglucan production by Agrobacterium tumefacien through orthogonal experiment and single-fac-tor test. [Result] The optimal medium consisted of 70 g/L sucrose,6 g/L yeast extract,10 g/L CaCO3,1. 5 g/L MgSO4 and 0. 025 g/L KH2PO4, and the optimal fermentation conditions included the incubation time of 4 d,inoculum amount of 2. 5%,initial pH of 7. 2,culture temperature of 30 ℃ and shaking speed of 250 r/min. Under these conditions,the yield of succinoglucan from Agrobacterium tumefacien was obviously improved to 18. 550 g/L,compared with the initial 8. 010 g/L. [Conclusion] The study can provide reliable basis for utilizing this Agrobacterium strain to make a further research into succinoglucan and to potentially produce succinoglucan in an industrial scale.
