摘    要:建立了二极管阵列检测器(DAD)测定红车轴草提取物中大豆黄素、染料木素、刺芒柄花素和鹰嘴豆芽素A的反相高效液相色谱分析方法,结果表明:大豆黄素、染料木素、刺芒柄花素和鹰嘴豆芽素A分别在0.0068~0.027、0.0145~0.058、0.021~0.084、0.008~0.032mg/ml与峰面积呈良好线性关系,加样回收率分别为96.3%、99.2%、100.1%、102.9%,RSD分别为3.7%、4.8%、4.3%、1.6%。该方法准确、快速、稳定、重现性好。

Abstract:A method was established for the identification and quantification of Daidzein,Genistein,Formononetin and Biochanin A in the extracts from Trifolium pratense with the reversed-phase liquid chromatography with an ultraviolet-diode array detector. The column was DiamonsilTM C18 column,250×4.6 mm and 5 μm particle. The solvent A was CH3CN; solvent B,0.1 % H3PO4,and the percentage of solvent A increased linearly,after an initial hold of 1 min,from 15 % to100 % in 60 min. The detection was by absorbance at 260 nm with PDA100 diode-array detector,thermostatted at 20 °C; elution with a binary high pressure gradient at a flow rate of 0.8 ml/min. The results showed there were good linearity correlations between the contents of Daidzein,Genistein,Formononetin and Biochanin A and their peak area was in 0.006 8~0.027,0.014 5~0.058,0.021~0.084 and 0.008~0.032 mg/mL respectively,the recovery capacity for each was 96.3 %,99.2 %,100.1 % and 102.9 %,and the RSD was 3.7 %,4.8 %,4.3 % and 1.6 %. The method established was accurate,fast,stable and the repeatability.
