玉米基因组DNA快速提取方法

摘    要:探讨了用NaOH裂解法从玉米单子粒胚乳中和用十二烷基肌氨酸钠法从玉米叶片中快速提取基因组DNA的方法,分析了2种方法提取的基因组DNA的效果及应用价值.结果表明,虽然得到的DNA质量基本一致,都可用于玉米品种纯度检验和分子标记辅助选择,但优化的、从叶片中快速提取的基因组DNA可长期保存,并用于多种后续序列分析工作.

Abstract:The effect and potential utilization of genomic DNA extracted from endosperm by NaOH method and leaf by SLS method were studied.The results showed that the quality of genomic DNA extracted using the two methods was almost consistent and could be used for purity test and marker assi-sted selection of maize.However,the genomic DNA extracted wtih SLS method could be conserved for a longel time and used in subsequent sequencing works.
