摘 要: | [目的]建立一种快速、简便、灵敏的堆肥中土霉素和金霉素残留的检测方法。[方法]采用高效液相色谱(HPLC)荧光检测法检测堆肥中的土霉素和金霉素残留,以V(咪唑缓冲溶液)∶V(甲醇)=80∶20(pH=5)为流动相,C18柱分离,用苯乙烯二乙烯苯共聚物小柱富集,净化。[结果]土霉素和金霉素的检出限分别为0.02、0.04mg/L;土霉素加标量为0.02、0.05、0.10mg/L时的回收率为78.0%~91.5%,方法的RSD为2.1%~7.9%,金霉素加标量为0.04、0.10、0.20mg/L时的回收率为87.5%~93.5%,方法的RSD为4.3%~6.5%。[结论]采用高效液相色谱荧光检测法检测堆肥中的土霉素和金霉素具有灵敏度高、稳定性好、简便易行等优点,且被测组分可得到较好的分离。

Abstract: | [Objective]The aim was to set up a rapid,simple and sensitive detection method for oxytetracycline and chlortetracycline residues in compost.[Method]The oxytetracycline and chlortetracycline residues in compost were detected by using HPLC-fluorescence detection method.The ratio of V(glyoxalin buffer solution)to V(methanol)of mobile phase was 80∶20(pH=5),the column C18 was used for separation and the styrene-divinyl-benzene copolymer columella was used for enrichment and purification.[Result]The detection limits of oxytetracycline and chlortetracycline were 0.02 and 0.04 mg/L resp.When the addition amount of standard oxytetracycline was 0.02,0.05 and 0.10 mg/L resp.,the recovery and RSD of this method were 78.0%-91.5% and 2.1%-7.9% resp.When the addition amount of standard chlortetracycline was 0.04,0.10 and 0.20 mg/L resp.,the recovery and RSD of this method were 87.5%-93.5% and 4.3%-6.5% resp.[Conclusion]The HPLC-fluorescence detection on oxytetracycline and chlortetracycline in compost had advantages of high sensitivity,good stability,simple,convenient and easy to operate and the detected components were separated better. |
