摘    要:为建立 Folin-Ciocalteu法测定大熊猫主食竹缺苞箭竹(Fargesia denudata)总酚酸含量的方法。以没食子酸为对照品,研究了Folin-Ciocalteu法测定缺苞箭竹中酚酸类物质含量的适宜条件。结果表明,适宜的比色条件为Folin-Ciocalteu试剂与 10% Na2CO3体积比为1∶6,Folin-Ciocalteu试剂3 mL,显色温度25℃,反应时间2 h,检测波长780 nm。浓度为20~100 μg·mL-1时与其吸光值呈良好的线性关系,测定方法的平均回收率为99.4%。可见该法有较好的准确度和重现性。

Abstract:Aims: to establish a quantitative method of measuring total phenols in Fargesia denudata, the main food of giant panda. Methods: with gallic acid as reference substance to study the suitable conditions of the measurement by Folin-Ciocalteu method. Results: the optimum conditions for determining total polyphenols were as follows: the volume ratio of Folin-Ciocalteu reagent to 10% Na2CO3: 1∶6, 3 mL of Folin -Ciocalteu reagent incubated at 25℃for 2 h, followed by detection at 780 nm. Good linear relationship was found in the concentration of 20-100 μg·mL-1, the average recovery was 99.4%.
