| 摘 要： | 为研究奶牛S100A12蛋白功能提供一种灵敏、高效的免疫学检测试剂,将纯化好的S100A12蛋白分别与弗氏完全佐剂和弗氏不完全佐剂乳化制备成抗原,免疫新西兰大白兔制备S100A12多克隆抗血清,应用琼脂双扩散法、间接ELISA法和Western blot方法检测该抗体的效价及其特异性。结果表明,所制备的S100A12抗血清经琼扩法和间接ELISA法检测效价分别达到1∶8和1∶409 600,同时免疫印迹法证实该抗体能与S100A12蛋白特异性结合。本试验成功获得了特异性强、效价高的S100A12多克隆抗体,为进一步深入研究S100A12基因功能提供了有力工具。

| Abstract: | To develop highly effective and sensitive antisera for further investigation on the functional of dairy cow S100A12 protein,emulsifying the purified S100A12 protein with the Freund’s complete adjuvant and Freund’s incomplete adjuvant respectively as antigen,rabbits were immunized with the antigen to obtain the polyclonal antisera.The titer was then tested using agar diffusion and indirect enzyme-linked immunosorbent assay(ELISA),and the specificity was tested by Western blot.The titer of the S100A12 was 1∶8 as determined by agar diffusion and over 1∶409 600 by ELISA,and it could specifically recognize S100A12 protein by Western blot.The results indicated that the high titer and specificity of anti-S100A12 polyclonal antibodies were obtained and it could be a novel tool in future for investigation of S100A12 functions. |
