去势雌性大鼠骨计量学研究

摘    要:切除3月龄SD大鼠双侧卵巢建立骨质疏松症动物模型,用图象定量法,将去势7周后的大鼠胫骨近端骨小梁测量,所得的各项参数以对照组比较的结果显示:去势组大鼠骨小梁面积减小+44%,周长-34%、密度-41%、间隙+99%、骨小梁的骨转换增加,如:四环素标记周长+77%、吸收陷窝周长+298%、矿化沉积率+32%、骨形成率+130%,但去卵巢大鼠骨吸收的增加超过骨形成的增加以导致骨质丢失和骨形态结构的改变。

Abstract:Female Sprague Dawley rats,at the age of 3 month were subjected to bilateral ovariectomy so as to set up an animal model for post-menopausal osteoporosis study . All rats were sacrificed at 7 weeks postovarectomy and their proximal tibiae were processed undecalcified for quantitativebone histomorphometry study with stereological method. In comparison to sham operatedcontrol rats, the proximal tibia of ovariectomized rats were characterized by significant decreased in cancellous bone volume (- 44%),trabecular perimeter (-34%), trabecular number ( -41%), trabecular separation(+99%),and increased in indices of bone turnover i. e. label perimeter(+ 77%),erded perimeter ( + 2.98%), mineral apposition rate ( + 32%), bone resorption of ovariectomized rate ( + 130%). In the final analysis, bone resorption ofovariectomized rats was more than formation, resulted in deprivation on bone mass and alterationof bone structure in ovariectomized rats.
