菠萝种质鉴定及亲缘关系的AFLP分析

摘    要:从FISH-AFLP试剂盒(PSTI/MseI型)64对引物中,筛选出8对AFLP选择性引物P-GAA/M-CTA、P-GAC/M-CTG、P-GAC/M-CTT、P-GAG/M-CAT、P-GAG/M-CTA、P-GAT/M-CAT、P-GAT/M-CTA、P-GAT/M-CTC,对39份菠萝种质进行了AFLP分析,都得到了清晰的多态性指纹图谱。8对引物共获得454个遗传位点,其中多态性位点332个,多态性比例平均为73.1%。UPGMA聚类表明,各种质间的相似系数为0.73~0.98,说明39份菠萝种质遗传关系相对较近。依相似系数0.80的水平,将供试的39份菠萝种质分为4类。研究结果可以作为菠萝分类的理论依据和基础。

Abstract:Eight pairs of primers, including P-GAA/M-CTA, P-GAC/M-CTG, P-GAC/M-CTT, P-GAG/M-CAT, P-GAG/M-CTA, P-GAT/M-CAT, P-GAT/M-CTA and P-GAT/M-CTC were selected for AFLP analysis from the 64 primer pairs supplied with FISH-AFLP kit. A total of 454 loci were amplified from 39 pineapple accessions, among which, 332 (73.1%) were polymorphic. UPGMA clustering analysis indicated that the Dice’s coefficient of the 39 pineapple accessions ranged from 0.73 to 0.98 indicating relatively close relationship among these accessions. Based on the results, the 39 accessions could be divided into 4 categories at a Dice coefficient value of 0.80. The results provided theoretical basis for the classification of pineapple.
