SaNHX耐盐基因转化红麻T_1代的耐盐性初步鉴定

摘    要:为鉴定转导SaNHX耐盐基因T1代980个红麻株系的耐盐性,本研究对980个株系在福州进行耐盐性鉴定和筛选,并对筛选出的9个耐盐T1代株系进行PCR检测和不同盐浓度水培和盆栽初步鉴定。通过调查转基因T1代在盐胁迫条件下的发芽率、出苗率、幼苗生长情况,对转基因后代的耐盐水平进行评价。结果表明:(1)转耐盐基因红麻T1代9个株系在0.71%的处理下,种子发芽率和生长势明显优于未转导对照红麻。苗高、苗重和根长分别比对照提高了2.04cm,0.23g和3.7cm。(2)0.3%、0.5%和0.71%三个不同盐浓度胁迫5天、10天、15天三个处理试验的方差分析表明,耐盐转基因T1代的株系幼苗成活率明显高于普通对照,差异均达极显著水平,其中编号176株系最佳;(3)在0.71%盐胁迫处理下,生长10天的T1代红麻幼苗苗高、苗重和根长与对照存在显著差异。在9个转基因株系中,176号植株最好,其苗高、苗重和根长的增长量分别比对照提高5.47cm、0.72g和4.54cm。

Abstract:To identify the salt-resistance level of 980 T1 plants of salt-tolerant SaNHX gene transformed kenaf,different salt endurance screening researches were conducted mainly in Fuzhou.Then the detection by PCR,different salt concentration solution cultures and pot experiments were carried out using 9 salt-tolerant T1 plants selected.Seed germination rate,emergence rate and seedling growth situation were investigated for T1 generation under salt-stressed conditions.The results showed that:(1) Seed germination rates of 9 T1 plants were obviously higher than the control and the growth status was obviously better than the control under 0.71% NaCl concentration treatment.The increases in seedling height,seedling weight and root length were 2.04cm,0.23g and 3.7cm respectively,compared to the control.(2) Under three different salt concentration stress of 0.3%、0.5%and 0.71% and three treatment experiment time of 5 days,10days,15 days,significant differences in the seeding survival rates were found through variance analysis,indicating that the seeding survival rate of transgenic T1 plants was higher than that of the control obviously,and the seedling survival rate of line 176 was the highest.(3)Under 0.71% salt stress treatment,there were significant differences statistically in the seedling height,seedling weight and root length of T1 generation plants of kenaf compared to the control after 10 days growth.In these 9 transgenic lines,line 176 was the best,and its seedling height,seedling weight and root length were increased by 5.47cm,0.72g and 4.54cm respectively compared to those of the control.
