摘    要:【目的】研究磷脂氢谷胱甘肽过氧化物酶(PHGPx)在山羊各种组织中的表达特点及在性腺中的定位情况。【方法】提取各种组织总RNA,应用实时荧光定量PCR方法检测各种组织PHGPx mRNA表达规律,利用免疫组化技术对睾丸和附睾PHGPx进行定位分析。【结果】PHGPx mRNA在各种组织表达水平由高到低依次为:睾丸心脑附睾肾肝肺脾背最长肌,在性腺和附睾中:睾丸附睾尾附睾头附睾体;免疫组化结果显示睾丸间质组织、生精细胞均有阳性产物,附睾头部平滑肌纤维表达弱阳性,附睾尾部附睾管中的单层柱状上皮表达呈强阳性。【结论】PHGPx在不同组织广泛表达,发挥重要的生物学功能,免疫组化结果直观显示了睾丸及附睾表达差异的部位,需深入研究其作用机制。

Abstract:【Objective】 The objectives of this study were to evaluate the expression pattern of PHGPx in various tissues and localization of PHGPx in gonad of goat. 【Method】 Total RNA was isolated from various tissues, and mRNA expression was detected by real-time fluorescent quantitative PCR. Cellular localization of PHGPx in testes and epididymidis was examined by immunohistochemistry, respectively. 【Result】 The quantitative real-time PCR results showed that the rank order of PHGPx mRNA expression was testes>>heart>cerebrum>epididymidis>kidney>liver>lung>spleen>Longissimus dorsi (LD) muscle, and testes>>epididymidis tail (EpT)>epididymidis head (EpH)>epididymidis body (EpB). There was a positive expression of PHGPx in interstitial tissue and spermatogenic cells of caprine testes. The expression of PHGPx in smooth muscle fibers (SMF) of EpB showed a weak positive signal, and simple columnar epithelium (SCE) around the ductus epididymidis (DE) of EpT showed a strong positive products. 【Conclusion】 Goat PHGPx mRNA appeared to be ubiquitously expressed almost in all tissues, which plays an important biological function. The immunohistochemistry results display the differently expressed region evidently between testes and epididymidis. Furthermore, the mechanism of dramaticly expressed in gonad needs to be further researched.
