鞘氨醇和水分胁迫对紫花苜蓿愈伤组织培养的影响

摘    要:研究了低于0.1 mg/L的不同浓度鞘氨醇处理对苜蓿子叶愈伤组织诱导及在干旱胁迫中的影响。结果表明,在15 d内该浓度范围的鞘氨醇明显抑制苜蓿愈伤组织形成,但30 d时,经鞘氨醇处理的愈伤组织诱导率与对照的无明显差异。继代培养时,供试不同浓度的鞘氨醇均明显降低细胞抗坏血酸过氧化物酶(APX)和过氧化物酶(POD)活性,但0.06 mg/L的鞘氨醇处理显著提高了POD的活性,在处理后15 d POD活性比对照增加了约2倍;不同浓度的鞘氨醇均使过氧化氢酶(CAT)活性大幅度升高,0.02 mg/L处理后15 d CAT活性增加了约8倍。在不同浓度聚乙二醇(PEG)胁迫下,鞘氨醇处理后紫花苜蓿愈伤组织中的过氧化物酶活性有所升高。

Abstract:Effects of the different concentrations of sphingosine below 0.1 mg/L on the induction of the callus and the drought stress of Medicago sativa L.were investigated.The experiment results showed that this range of concentration of sphingosine have no effect on the induction of the callus,and in 15 d the induction rates of the treated callus are lower than that of the check.control but in 30 d,there are not wide gaps between the induction rates of the treated callus with sphingosine and that of the check.Control group during the subculturing,sphingosine reduced the activity of POD and APX,but the treatment of 0.06 mg/L sphingosine promoted the activity of POD after 15 days,the activity increased by about 200%;and different concentrations of sphingosine promoted the activity of CAT by a big margin.The treatment of 0.02 mg/L sphingosine,after 15 days,the activity of CAT increased by about 800%.We treated callus of alfalfa with spingosine,and water-stressed using polyethylene glycol(PEG) as the stress reagent,the result showed that the activity of peroxides were higher than the untreated.
