摘    要:以杉木实生苗的子叶和下胚轴为起始外植体,研究了基本培养基、6-BA、TDZ、苗龄等因素对器官发生和体胚发生的影响.研究结果表明:基本培养基、6-BA、TDZ及苗龄对子叶和下胚轴不定芽发生和体胚发生均产生显著影响.DCR+6-BA 1.0mg/L+TDZ 0.003mg/L+NAA 0.1mg/L是子叶和下胚轴诱导不定芽发生及下胚轴诱导体胚发生的最佳培养基;DCR+6-BA 1.0mg/L+TDZ 0.002mg/L+NAA 0.1mg/L对子叶诱导体胚最有效;不定芽在DCR基本培养基中可有效伸长;1/4MS+IBA 0.2mg/L+NAA 0.1mg/L可有效诱导不定芽生根.

Abstract:Using cotyledons and hypocotyls of seedlings as initial explants, we s tudied the effect of such factors as basal medium, 6-BA,TDZ and seedling age on organogenesis and somatic embryogenesis. The results indicated that basal mediu m, 6-BA, TDZ and seedling age greatly influenced the induction frequency of orga nogenesis and somatic embryogenesis. The medium of DCR+6-BA 1.0mg/L + TDZ 0.003m g/L+NAA 0.1 mg/L was optimal for cotyledons and hypocotyls to induce adventitiou s buds and hypocotyls to induce somatic embryos. The optimal medium for cotyled ons to induce somatic embryos was DCR+6-BA 1.0mg/L+TDZ 0.002 mg/L+ NAA 0.1 mg/L. Adventitious buds can elongate effectively on the DCR medium without plant horm one. 1/4MS+IBA 0.2 mg/L +NAA 0.1 mg/L was the optimal medium for adventitious bu ds to root.
