谷胱甘肽与顺铂联用对肺癌A549细胞增殖的影响

目的探讨谷胱甘肽与顺铂联用对肺癌A549细胞增殖的影响。方法应用MTT法及细胞计数评价细胞增殖情况,流式细胞术检测细胞凋亡及活性氧(ROS)水平,Western blot检测P53表达。结果 0.3、0.9g/L谷胱甘肽与3mg/L顺铂联用72h,1、3、9g/L谷胱甘肽与9mg/L顺铂联用30h,均可显著降低顺铂对A549细胞增殖的抑制作用(P<0.01)。9g/L谷胱甘肽与9mg/L顺铂联用,可减少A549细胞ROS产生和早期凋亡(P<0.01)。谷胱甘肽、顺铂单独或联用对P53蛋白表达无影响。结论谷胱甘肽与顺铂联用对A549增殖的影响取决于谷胱甘肽剂量。

Effect of glutathione and cisplatin on lung cancer A549 Cells

Objective To investigate the effect of glutathione and cisplatin on proliferation of lung cancer A549 cells. Methods Cell proliferation, apoptosis, reactive oxygen species （ROS） and P53 expression were evaluated by MTT, cell counting, flow cytometry and Western blot, respectively. Results Cisplatin-induced inhibition of A549 cell proliferation decreased remarkably （P〈0.01） after coculture of 0.3 g/L or 0.9 g/L glutathione and 3 mg/L cisplatin for 72 h, or 1, 3,9 g/L glutathione and 9 mg/L cisplatin for 30 h. Cotreatment of 9 g/L glutathione and 9 mg/L cisplatin inhibited ROS production and early apoptosis of A549 cells （P〈0.01）. Single or combined use of glutathione and cisplatin had no impact on P53 expression of A549 cells. Conclusion The doses of glutathione depend on the effect of glutathione and cisplatin on A549 cells.
