花叶夹竹桃离体培养技术研究

摘    要:通过正交试验等方法对花叶夹竹桃茎段组织培养技术进行了研究.结果表明:在江苏句容,花叶夹竹桃最佳取材时间为4月下旬.在优良母株上选取外植体,经过0.1%HgCl2消毒15min,灭菌成活率为78.8%;添加3.5%的白砂糖和增加培养容器的透气性,能很好地预防玻璃苗的发生;改良MS附加ZT,BA,NAA等植物生长调节剂可以有效地促进试管芽苗的增殖和生长,其中ZT1.5mg/L+BA2.0mg/L十IBA0.1mg/L组合最适宜增殖与生长培养,增殖达5.6倍,生长高度为3.2cm;适宜的生根培养基为1/2MS+IBA0.5mg/L+NAA0.3mg/L+Charcoal3000mg/L+白砂糖2%,生根率可达92%;经生根的试管苗移栽于V(蛭石):V(珍珠岩):V(泥炭)为6:3:1的混合基质中,控制温度在23~30℃,相对湿度90%,保湿12~18d,其间适当遮荫,30d后成活率达88%以上。

Stem Segment Culture in Vitro of Nerium indicum cv. Variegata

Abstract:This experiment concerning Nerium indicum cv. Variegata. segment culture in vitro was committed on the explants sterilization,vitreous shoots control, shoot multiplication and root induction. The results showed that: The latter of April was optimal collection time in vitro. After 0.1% HgCl2 sterilization for 15 min, the survival rate for the forthcoming culture accounted up to 78.8% ;addition of 3.5% sugar and improvement on air-permeation of culture containers could effectively control vitreous shoots;the modified MS media supplemented with ZT, BA and NAA could ameliorate the culture multiplication and growth, among which the combination of 1.5 mg/L ZT + 2.0 mg/L BA + 0.1 mg/L IBA was optimal, with the multiplication time was up to 5.6 and growth height up to 3.2 cm ; the rooting medium 1/2 MS + 0.5 mg/L IBA + 0.3 mg/L NAA + 3 000 mg/L Charcoal + 2% sugar was optimal,rooting percentage was up to 92% ;the root-developed plantlets transplanted in the substrate mixed with vermiculite, pearlite and peat( volumn ratio of 6 : 3 : 1 ) at 23 - 30 ℃, relative humidity was 90% for 12 - 1 g d, favorably shaded,the survival rate could get over 88% after 30 d.
