甜菜未授粉胚珠培养技术研究

摘    要:甜菜未授粉胚珠接种在附加不同生长素(IAA、2,4—D)、细胞分裂素(KT、6BA)或激素组合(6BA NAA和IAA KT)的MS、PGOB、1/2MS等培养基上,可成功诱导出胚状体,分化诱导形成单倍体植株,对其用0.1%~03%秋水仙碱溶液处理使染色体加倍,获得了高度纯合的二倍体株系。试验选配了110种诱导培养基、两种分化和3种生根培养基,选择了143种不同基因型材料.不同基因型材料在不同培养基上的诱导率各不相同.

Abstract:Results showed that unpollinated ovules, when inoculated on MS, PGOB, 1/2 MS media with various auxin (IAA or 2,4-D), cytokinins (KT or 6BA) or hormone combinations (6BA NAA and IAA K) added in, could be induced to produce embryoids which could be further induced to differentiate and to form haploid plants. Highly homozygous diploid lines were obtained from these haploids treated with 0.1%-0.3% colchicine solution to give chromosomes doubling. In this experiment, 110 induction media, 3 rooting producing media and 2 differential media were utilized in combination with 143 genotypes. The percentage for inducing embryoids and the resulting haploids were not same with different genotype on various media.
