白魔芋种质资源的RAPD分析

摘    要:白魔芋原产于中国,是魔芋属中品质最优的种,种内资源类型丰富,为进一步分析各类型或产地间的亲缘关系,应用RAPD分子标记技术,用40个随机引物(10-mer)对34份白魔芋种内资源基因组DNA进行PCR扩增,有11条引物能够在34份材料中扩增出112条稳定的带,其中有93呈多态性。用dps软件对扩增产物进行聚类分析,根据聚类结果将34份材料划分为5类,通过与叶柄斑纹比较分析,发现各类与叶柄斑纹关系不紧密,而有较为明显的地域相关性,该结果为深入了解白魔芋种内资源的亲缘关系,提供了分子水平的依据

Abstract:Amorphophallus albus native to China is one of the precious wild species in the genus Amorphophallus. The rich genetic variations were also found within the species. Using the RAPD techniques, the phylogenetic relationships among the different bio-types or origin places of the species were analyzed in the present study. The genomic DNAs isolated from 34 accessions of A. albus were amplified by using 40 decamer primers. Eleven primers showed consistent banding patterns and amplifications in all the materials tested and produced 112 bands, of which 93 bonds were polymorphic. Cluster analysis using dps program shows that the 43 materials were divided into 5 groups. A comparison of this result with the leaf-stem morphology, we found that the clusters were highly correlated with the geographic origins of the materials. This result provided molecular information for a better understanding of the genetic relationship of A.albus species.
