摘 要: | [目的]从新疆五家渠市耐盐碱植物盐穗木根际土壤中分离出兼性耐盐菌B6,并对其分别进行形态特征、生理生化特性分析及16S rDNA鉴定。[方法]采用真细菌16S rDNA通用引物PCR扩增B6菌株的16S rDNA,使用NCBI-Blast软件在GenBank数据库中对其进行同源性检索,使用MEGA4软件构建系统发育树。[结果]B6菌能在0~20%NaCl的条件下生长,为有芽孢的杆状菌;B6的16S rD-NA长度为1 446 bp,与多种地衣芽孢杆菌的16S rDNA序列同源性高达99%。[结论]系统发育树表明B6菌株与地衣芽孢杆菌的亲缘关系最近,结合生理生化分析,可判断B6菌株为地衣芽孢杆菌。

Isolation and Identification of A Halotolerant Bacteria Strain B6 Isolated from Rhizosphere Microbe of Halostachys Caspica |

Abstract: | The common primers for the 16S rDNA of eubacteria were used for polymerase chain reaction(PCR). Homology analysis was made by searching in Genbank with NCBI-Blast and the phylogenetic tree was constructed with MEGA4. [ Result] Strain B6 can grow on the media containing 0 -20% NaC1, and was bacilli with spore forming. The amplified 16S rDNA fragment length was 1 446 bp and the result indicated that the 16S rDNA homologies were 99% between strain B6 and many kinds of Bacillus Licheniformises. [ Conclusion] The tree showed the clo- sest relationship was between strain B6 and Bacillus licheniformis. By analyzing the physio-biochemical characteristics of strain B6, strain B6 could be identified as a species of Bacillus licheniformis. |
