摘    要:目的]测定新疆产不同品种葡萄干中有机酸的含量。方法]采用Symmetry ShieldTMRP18(4.6 mm×250 mm,5μm)色谱柱,以磷酸盐缓冲溶液(pH 3.0)为流动相,流速1.0 ml/min,柱温30℃,紫外检测波长210 nm,建立了葡萄干中酒石酸、苹果酸等4种有机酸的高效液相色谱定量分析方法,测定了新疆8种不同品种葡萄干中的酒石酸、苹果酸、柠檬酸和丁二酸的含量。结果]该方法相对标准偏差(RSD)0.57%~1.78%,回收率97.6%~101.2%,各种酸的线性相关系数r>0.999 7。结论]所建立的方法快速、灵敏度高、重复性好,可用于葡萄干中有机酸的测定。

Determination of Organic Acids Content in Different Varieties of Raisin in Xinjiang by HPLC

Abstract:Objective] To determine organic acid content in different varieties of raisin in Xinjiang.Method] By using Symmetry ShieldTM RP18(4.6 mm × 250 mm,5 μm) column,with phosphate buffer solution(pH 3.0) as the mobile phase at a flow rate of 1.0 ml/min,column temperature 30 ℃,UV detection wavelength 210 nm,a quantitative reversed-phase high performance liquid chromatography(HPLC) method was established for the determination of tartaric acid and malic acid in raisin.The contents of tartaric acid,malic acid,citric acid,succinic acid in 8 varieties of raisin in Xinjiang were determined.Result] RSD is 0.57%-1.78%,recovery rate 97.6%-101.2%,linear correlation coefficient of each acid is r0.999 7.Conclusion] The method could be applied in determination of organic acids in grape with characteristics of rapid,good sensitive and repeatability.
