摘 要: | [目的]探讨五味子甲素、乙素及丙素对人皮肤细胞衰老作用的影响,为开发北五味子抗皮肤衰老产品奠定基础。[方法]通过H2O2造成HaCat细胞氧化损伤衰老模型,采用噻唑蓝(MTT)法测定HaCat细胞增殖情况。[结果]浓度50μmol/L的H2O2对细胞无明显抑制作用,浓度100和200μmol/L组有明显抑制作用。H2O2组细胞的存活率为(71.667±0.862)%。浓度100和200μmol/L五味子甲素组细胞的存活率分别为(76.400±0.656)%和(82.367±0.666)%;浓度100和200μmol/L五味子乙素组细胞的存活率分别为(86.300±0.721)%和(88.900±0.985)%;浓度100和200μmol/L五味子丙素组细胞的存活率分别为(86.767±0.551)%和(88.067±0.873)%。[结论]五味子甲素、乙素及丙素对H2O2造成细胞的氧化损伤均具有一定的保护作用,以五味子乙素和丙素的效果较好。

Abstract: | [ Objective ] To research the protective effects of schisandrin A, B and C on the oxidative damage of HaCat cells, so as to provide foundations for the development of schisandrin anti - aging products. [ Method] Aging model was established based on the oxidative damage of HaCat ceUs induced by H2O2. And proliferation of HaCat cells was detected by MTY assay. [ Result] 50 μmol/L H2O2 showed no inhibitory effects on cells; but 100 and 200 μmol/L groups had significant inhibitory effects. The survival rate of cells in H202 group was (71. 667 ± 0. 862) %. The survival rates of cells in 100 and 200 μmol/L schisandrin A groups were (76.400 ± 0.656 )% and (82. 367 ± 0.666), re- spectively. The survival rates of cells in 100 and 200 μmol/L schisandrin B groups were ( 86.300 ± 0.721 ) % and ( 88.900 ± 0.985 ) % , re- spectively. The survival rates of cells in 100 and 200 μmol/L schisandrin C groups were ( 86.767 ± 0.551 ) % and ( 88. 067 ± 0. 873) %, re- spectively. [ Conclusion] Schisandrin A, B and C all had certain protective effects on the oxidative damage of HaCat cells induced by H202. And achisandrin B and C showed relatively better effects. |
