甘薯二羟基黄酮醇还原酶基因的克隆及在大肠杆菌中的表达

摘    要:根据其它作物的二羟基黄酮醇还原酶(DFR)基因的保守序列设计引物,利用RT-PCR方法从甘薯块根中分离出花青素合成相关基因DFR基因,并定名为ibDFR。该基因全长1232bp,编码398aa。核甘酸序列分析表明,与牵牛相似系数达85%,与烟草的相似系数达到67%,与马铃薯的相似系数达75%。将ibDFR连接到pET30a(+)载体中,检测ibDFR在大肠杆菌中的表达。实验结果表明ibDFR蛋白质分子量大小与推算一致,能在大肠杆菌中转录表达。

Abstract:According to the conserved sequences of dihydroflavonol 4-reductase (DFR) genes from other plants, primers were designed to amplify DFR gene from sweet potato, and the gene was deginated ibDFR in this paper. The gene have 1 232 bp necleounide and encoded 398 aa. Sequence comparison revealed that ibDFR sequence shared homologies of 85%, 67% and 75% with those of Ipomoea purpurea, tobacco and potato.
