柳叶蜡梅叶片愈伤组织诱导实验研究

摘    要:以不同方式处理柳叶蜡梅种子,观察其在MS培养基上萌发和生长的状况。结果显示:98%浓硫酸处理的种子发芽率达32%,其它处理萌发率低。以种子无菌苗叶为外植体,接种于2,4-D、NAA、6-BA组合不同浓度生长调节剂的MS固体培养基上,进行愈伤组织诱导实验;外植体愈伤组织诱导培养基MS+NAA0.3mg·L^-1+6-BA1.0mg·L^-1的诱导率为80%,生长稳定可分化出苗。

Abstract:Seeds of Chirnonanthus salicifolius by different treatments were cultured on solid MS medium. Results showed that the germination rate reached 32 % after the seeds were soaked in 98% sulfuric acid and the other treatments had a low termination rate. Explants from the aseptic leaves were incubated on solid MS medium supplemented with 2,4-D, NAA, 6-BA or different combinations of these plant growth regulators to induce callus. The formula of MS+NAA 0.3mg· L^-1 +6-BA 1.0mg · L^-1 had the best result, with the induction rate of 80 G and plantlets were differentiated afterwards.
