摘    要:目的]将YB2基因导入玉米自交系.方法]利用花粉管通道法将含有YB2基因的线性表达盒转入优良玉米自交系京24,对T0代植物喷洒浓度为200 mg/L草铵膦药液进行筛选,并对筛选到的植株进行PCR、RT-PCR检测.结果]试验共得到19株草铵膦抗性植株;经检测,19株均为Bar阳性植物,其中5株含YB2目的基因,初步证明外源目的基因已整合到玉米基因组中.结论]该研究为获得抗旱性好、产量高、安全的转基因玉米株系奠定了基础.

Study on the Transferring Linear of YB2 Gene into Maize Inbred Lines via Pollen Tube Pathway

Abstract: Objective/ The aim was to introduce Yb'2 gene into maize inbred hnes. Method J lanear expressing cassette commnmg YB2 gene was transferred into maize inbred line Jing 24 by pollen tube pathway method. To generation plants were screened by spraying 200 mg/L glufosi- nate herbicide,and the screened plants were detected by PCR and RT-PCR. ~ Result] 19 glufosinate-tolerant plants were obtained. All of them were Bar positive plants ,5 of them contained YB2 gene ,which proved that exogenous gene had been integrated into the genome of maize. Con- clusion ] The study laid a foundation for obtaining transgenic maize lines with good drought resistance, high yield and good security.
