台湾青枣不同品种表型差异和遗传多样性研究

摘    要:对台湾青枣9个品种的15个形态指标进行了统计分析,同时使用ISSR分子标记技术对其进行了基因组DNA多态性分析,从而确定台湾青枣形态特征与遗传差异的关系。结果表明:这些形态指标均达到明显的差异。用筛选出的10条能获得多态性高、重复性好、清晰位点的引物对100份DNA样品进行PCR扩增,每个引物可以扩增出3~8条带,这些DNA片段大小主要集中在250~2000bp,从明显的多态性位点可以得知该品种种质资源之间遗传多样性很低,同时也说明了台湾青枣的不同品种是可以用分子标记的技术来分析和鉴定的。

Study on the Phenotypic Difference and Genetic Diversity of Different Cultivars of Taiwan Green Jujube

Abstract:The study makes a statistical analysis for 15 morphological indexs of 9 different cultivars of Taiwan green jujube, and inter-simple sequence repeat (ISSR) method was used in analyzing the genetic diversity of these cultivars to determine the relationship of morphological characteristics and genetic difference of Taiwan green jujube. The result showed that these morphological indexs were obvious differences. 100 DNA samples were amplified using ten random PCR primers of high reproducible,clear and stable quality which were screened from sixty primers, every primer had 3 to 8 bands most of the bands were from 250 to 2000 bp, it was concluded from obvious polymorphic loci that the genetic diversity was very low in different idioplasmic sources of green jujube, at the same time, different cultivars of Taiwan green jujube could be analyzed and identified by Molecular Marker Technique.
