不同地区毛鸡骨草与鸡骨草rbcL 序列差异分析

摘    要:应用改良CTAB法提取不同产地毛鸡骨草与鸡骨草的总DNA,PCR 扩增其rbcL 序列并测序,应用MEGE5.1 软件作 序列同源性分析,计算遗传距离并构建系统发育邻接树。结果表明,毛鸡骨草与鸡骨草各具为一支,有亲缘关系,可见rbcL 序列可成功区分两者,因此,rbcL 序列可以作为鸡骨草的DNA 条形码。

Analysis of rbcL sequence variations between Abrus rnollis and A. cantoniensis in different areas
Abstract:Total DNA of Abrus rnollis and A. cantoniensis in different areas was extracted by modified CTAB, PCR amplification of rbcL was sequenced, MEGE5.1 software was applied to take contrived sequence analysis, genetic distance was calculated, and phylogenetic adjacent tree was eatablished. The results showed that A. rnollis and A. cantoniensis reached a branch each with phylogenetic relationship, indicating rbcL sequence could successfully distinguish the both. Therefore, rbcL sequences can be used as DNA barcode of A. cantoniensis.
