花粉管通道法转化甜瓜自交系的技术研究初报

摘    要:利用花粉管通道技术转化3个甜瓜自交系,筛选适宜浓度卡那霉素Kan对转化后代进行早期快速检测。试验结果表明,微量注射法转化率高于柱头滴加,切去柱头二次滴加更有利于外源基因转化。3个自交系转化率为0~1.4%,品种之间存在差异,乌克兰、雪里华、鲁薄1号平均转化率分别为0.30%、0.23%、0.25%;利用Kan临界浓度对转化种子进行早期筛选是可行的,子叶是适宜的检测时期。通过对Kan抗性苗的PCR检测证明,花粉管通道法可以实现甜瓜的遗传转化。

Abstract:The experiments using pollen-tube pathway to transform three muskmelon inbred lines, and selecting suitable concentration of Kan to detect transformation plants early and quickly were carried out. The results showed that the transformation efficiency of microinjection was higher than that of stigma-dropping. Cutting stigma and dropping repeatly was profit for transformation of extraneous gene. The transformation efficiency of three muskmelon inbred lines was 0- 1.4% and there were differences between them. The average transformation efficiencies of TS1, TS2 and TS3 were 0.30%, 0. 225% and 0. 25%. It was feasible to detect transgenic seeds early by critical concentration of Kan, and the accuracy efficiency of detecting cotyledons was higher. PCR results of seed- lings resistant to Kan showed that transforming muskmelon by pollen-tube pathway was successful.
