6-BA浓度对桔梗胚珠愈伤组织分化率的影响

摘    要:[目的]研究6-BA浓度对桔梗胚珠愈伤组织分化率的影响.[方法]以桔梗未受精胚珠为外植体,利用N6 +1.0 mg/L2,4-D+0.1 mg/L KT培养基诱导出的愈伤组织作为接种材料,分别接在0.5 mg/L NAA与不同6-BA浓度的分化培养基上,对其愈伤组织分化率进行研究.[结果]6-BA浓度在0.25、1.25和1.00 mg/L(即3种分化培养基为N6+ 0.5 mg/L NAA+ 0.25 mg/L 6-BA、N6+ 0.5 mg/LNAA+ 1.25 mg/L 6-BA、N6 +0.5 mg/L NAA+1.00 mg/L6-BA)时的分化率高,分别为88.3%、85.0%和80.0%.[结论]6-BA浓度为0.25 mg/L时有利于桔梗未受精胚珠愈伤组织的分化.

Abstract:[ Objective ] To study the effect of different concentration of 6-BA on differentiation rate of ovule callus of Platycodon grandiflorum. [ Methods] Using callus induced from N6 + 1.0 mg/L 2,4-D + 0. 1 mg/L KT medium and inoculated on 0.5 mg/L NAA differentiation media contained different concentration of 6-BA with unfertilized ovtdes of Platycodon grandiflorum as explant. [ Results] The results showed that, the differentiation rate under the differentiation media of N6 +0.5 mg/L NAA +0.25 mg/L 6-BA, N6 +0.5 mg/L NAA + 1.25 mg/L 6-BA and N6 + 0.5 mg/L NAA + 1.00 mg/L 6-BA are higher, up to 88.3%, 85.0%, 80.0% respectively. [ Conclusion ] At 6-BA concentration of 0.25 mg/L, it is favorable for differentiation of unfertilized ovules of Platycodon grandiflorum.
