荞麦愈伤组织培养及其分化的研究

摘    要:以荞麦的成熟胚,幼胚,下胚轴为外植体,在不同激素浓度配比,不同基本培养基质培养基上的组织培养研究表明:以成熟胚为外植体时,对愈伤组织诱导和生长最适培养基是2.4-D2.0,NAA1.0,6-BA。0.5的B_5培养基;以幼胚为外植体时,对愈伤组织的诱导和生长最适合的培养基是2.4-D2.0,NAA1.0,6-BA0的MS培养基;以下胚轴为外植体时,B_5和MS两种培养基均不适合愈伤组织的生长。2.4-0对荞麦愈伤组织根的分化有一定的促进作用。

Abstract:Using mature embryo,young embryo,and hypocotyl of buckwheat asexplant,we studied the induction of callus under different concentrations of promoting substance and different kinds of basal medium.The results were as follows:If embryo of buckwheat was used as explant,the most suitable medium for the induction of callus was the B_5 basal medium supplemented with 2mg/12,4-D,1.0mg/1 NAA,0.5mg/16-BA;if young embry was used as explant,the most suitable medium for the induction of callus was the MS basal medium supplemented with 2mg/12.4-D,1.0mg/l NAA,0mg/1 6-BA;if hypocotyl was used as explant,the B_5 and MS basal medium was not suitable for induction of callus.
