摘    要:本实验利用16SrDNA序列分析技术,对来自犊牛腹泻粪便中的四株菌(A菌,B菌,C菌,D菌)进行菌种鉴定。使用16SrDNA通用引物,通过PCR扩增分别得到1500bp,与GenBank中序列进行比对。结果表明:A菌与肠杆菌属、B菌与短小芽孢杆菌、C菌与大肠杆菌、D菌与变形杆菌的同源性最高,相似率均达到了99%,初步确定A菌为肠杆菌属、B菌为短小芽孢杆菌、C菌为大肠杆菌、D菌为变形杆菌。本试验的目的是为16SrDNA在临床细菌的鉴定提供客观理论依据。

Abstract:This experiment used sequence analysis of 16SrDNA for four bacteria ( A, B, C, D) from calf diarrhea feces. The test used PCR, amplified fragment was 1 500 bp by 16S rDNA universal primers. The sequence blasted with Gene Bank. The results showed that: the A bacteria was the Enterobacter sp, the B bacteria was the Bacillus pumilus strain, the C bacteria was the Escheriehia coli and the D bacteria was the Proteus vulgaris, the similarity ratio was reached 99% , initially that the A bacteria identified the Enter- obacter sp, the B bacteria identified the Bacillus pumilus strain, the C bacteria identified the Escherichia coli and the D bacteria iden- tified the Proteus vulgaris. The experiment aimed to provided the theoretical basis of the identification clinical bacteria by 16S rDNA.
